Time-resolved confocal scanning Fluorescence Lifetime Imaging Microscope (Microtime 200, Picoquant), suitable for:
1) Fluorescence Lifetime Imaging (time domain), e.g. life-time FRET
2a) Fluorescence Correlation & Cross-Correlation Spectroscopy (FCS & FCCS)
2b) Fluorescence Lifetime Correlation & Cross-Correlation Spectroscopy (FLCS & FLCCS)
2c) Other spatial/temporal fluorescence correlation studies
3) Steady-state and Time-resolved Fluorescence Anisotropy (TRFA) - polarization sensivitive lifetime measurements
4) Photon Antibunching measurements
Specs:
- two hybrid PMA detectors
- 4 laser lines (rep rate 0-40MHz): 440nm, 485nm, 510nm, 561nm. Possibility of pulse interlieved (PIE) excitation
- Piezo Objective-scanning.
- Temperature control for live cell imaging (for sample & objective lens)
- 355nm kHz-picosecond laser for inducing e.g. DNA damage at selected areas
- Analysis software: Symphotime 64 (Picoquant), Fluofit (Picoquant), custom written software
