Das Transmissionselektronenmikroskop LEO 912 AB (Zeiss) mit integriertem Omega-Energiefilter dient der hochauflösenden Untersuchung von biologischen Strukturen vom Molekül bis hin zu Zellbestandteilen wie Organellen oder extrazelluläres Oberflächenstrukturen.
Je nach Anwendung kann das Gerät mit einer Beschleunigungsspannung zwischen 60 und 120 kV betrieben werden und ist mit einer 2K SlowScan Kamera ausgestattet. Als Elektronenquelle dient ein LaB6 Kristall.
Strukturelle Veränderungen im Entwicklungsablauf verschiedener pflanzlicher oder tierischer Zellen oder Organismen können dargestellt und Veränderungen nach experimenteller Beeinflussung sowie unter veränderten Umweltbedingungen (Stress) untersucht werden.
Eine Elektronentomographie-Einrichtung und die dazugehörige Software zur Aufnahme und Analyse von Bildserien (Kippserien von +/-70°) erlaubt 3-D Rekonstruktionen von Zellbestandteilen. Die Ausstattung mit einem Energiefilter (Spektrometer) und einer entsprechenden Software zur Aufnahme und Analyse von Spektren (ITEM, Olympus) ermöglicht Elementanalysen mithilfe von Energieverlustspektroskopie (EELS) und die Darstellung von Elementverteilungsbildern bei hoher Auflösung an Ultradünnschnitten.
Anwendungsbereiche dafür sind einerseits die biologische Grundlagenforschung, in der analytische Elektronenmikroskopie zur Detektion von Markern und Indikatoren eingesetzt wird, und andererseits umweltbezogene Forschung z.B. zur Auffindung und Lokalisierung von verschiedenen Umweltschadstoffen, wie Schwermetallen in sub-zellulären Kompartimenten oder Organellen.
